Lack of Pineal Melatonin Removes Daily Rhythm of Key Melatonin-Making Genes in Rat Testes

Updated

Abstract

Pinealectomy in Wistar rats abolished 24-hour rhythmicity in the mRNA expression of several key genes related to melatonin and steroid production.

  • Control rats exhibited daily rhythms in mRNA expression for melatonin-related genes, peaking during the dark phase.
  • Pinealectomy disrupted the normal patterns of mRNA expression for multiple genes, shifting peak times to the light phase.
  • Melatonin replacement therapy restored normal expression patterns for some genes, including Lhr, Rev-erbα, and Per1, but not for Mt1.
  • Significant daytime differences were observed for several steroidogenic and clock genes in control animals, indicating their potential regulation by circadian rhythms.
  • The findings suggest that gene expression related to melatonin is likely influenced by the presence of pineal melatonin.
