Improved Ionizable Lipids That Break Down Easily and Escape Cell Compartments Better for Delivering mRNA and tRNA

Updated

Abstract

Essence

Thioester-containing ionizable lipids improved RNA delivery performance and biodegradability in experimental models.

Evidence

This formulation and platform experiment synthesized a thioester-linked ionizable lipid library and used in vivo structure-activity studies to identify CP-LC-1272, whose LNPs improved mRNA expression versus SM-102 and achieved similar protein restoration in a tRNA premature stop-codon model.

Caveat

The abstract reports platform and model-system performance, with safety, repeated dosing, and high-dose therapeutic claims remaining inferred rather than clinically tested.
