How Substrate Access and Position Affect Cas12a Protein Cutting Speed

Updated

Abstract

Kinetic models quantitatively describe critical molecular interactions in Cas12a ribonucleoproteins.

  • The apparent enzyme turnover number (k* cat) and Michaelis constant (K* M) were determined experimentally.
  • Two key processes were identified: substrate passage through the gap between the recognition and nuclease domains, and substrate alignment with the catalytic site.
  • The rate of substrate passage (Kpass) and alignment (Kalign) were quantitatively assessed through rational design of substrates and activators.
  • Consistent predictions from kinetic models aligned with experimental results, highlighting the importance of these molecular interactions for CRISPR functions.
