Type IIB PKA mainly controls insulin release boosted by Gs-linked receptors in mice by managing ion channels and insulin granule types

Abstract

RIIβ-knockout mice exhibited impaired glucose tolerance and attenuated insulin secretion in response to liraglutide.

  • Type IIB protein kinase A (PKA) is identified as a key mediator in insulin secretion regulation by glucagon-like peptide-1 (GLP-1) signalling.
  • Isolated islets from RIIβ-knockout mice showed decreased insulin secretion following stimulation with liraglutide, glucagon, and follicle-stimulating hormone (FSH).
  • RIIβ deficiency impaired the activation of PKA signalling in beta cells, affecting insulin secretion dynamics.
  • RIIβ-ablated beta cells exhibited reduced activity of potassium channels and downregulation of multiple voltage-gated calcium channel genes.
  • Insulin granule characteristics such as size, number, and docking at the plasma membrane were adversely affected in RIIβ-knockout beta cells.
