TYR-deficient human iPSC-derived retinal pigment epithelium formed pre- but lacked mature melanosomes.

Evidence

This in vitro CRISPR-Cas9 knockout model compared isogenic control and TYR knockout human iPSC-derived RPE using electron microscopy, protein staining, morphology, junction markers, and transepithelial resistance.

Caveat

The findings validate a cell model of -like pigmentation defects but do not show in vivo retinal development or clinical outcomes.
