Abstract

The engineered ultrasound-activatable lipid nanoparticle formulation increased antigen-specific CD8T cell responses by 2.5-fold compared to conventional formulations.

  • Sono-adjuvant lipid was developed to provide targeted immune stimulation in lymph nodes.
  • Ultrasound activation of the lipid nanoparticles enhanced immune responses by activating specific cellular pathways in antigen-presenting cells.
  • The combination of ultrasound with the novel lipid nanoparticles improved lymph node-specific mRNA transfection.
  • The strategy effectively boosted antitumor T cell immunity and completely eliminated orthotopic liver tumors in mice.
