Abstract

Cue-induced reinstatement of cocaine seeking may depend on the activation of a small population of neurons expressing neuronal nitric oxide synthase (nNOS).

  • Relapse to drug use can be triggered by drug-associated cues, which correlate with increased synaptic strength in certain brain regions.
  • Transient synaptic potentiation at glutamate synapses is linked to the spillover of glutamate from prefrontal cortical inputs.
  • Activation of metabotropic glutamate receptor 5 (mGluR5) is necessary for the increase in nitric oxide (NO) production during cue-induced reinstatement.
  • Selective elimination of nNOS interneurons resulted in a reduced intensity of cue-induced reinstatement, suggesting their critical role in this process.
  • The stimulation of nNOS interneurons can activate matrix metalloproteinases, which are involved in synaptic changes associated with relapse.
