Finding Protein Phosphatases That Work in the Fruit Fly's Body Clock Using RNAi

Abstract

Nineteen protein phosphatases were identified that altered circadian activity rhythms by at least 1 hour.

  • The screen involved RNA interference knockdowns of all annotated protein phosphatases in Drosophila.
  • Alterations in circadian period were defined as lengthening or shortening of rhythms or being arrhythmic.
  • Fifteen of the identified protein phosphatases were validated for further studies.
  • These phosphatases may contribute to understanding the circadian timekeeping mechanism in Drosophila and potentially in other animals.
