Chronic Light Exposure Causes Body Clock Disruption Linked to Faster Metabolic Aging in Rats, Prevented and Reversed by Melatonin

Abstract

A 5.1-fold decrease in plasma melatonin levels was observed in rats exposed to constant illumination over 24 months.

  • Chronic exposure to constant light caused significant metabolic disturbances, including hypertriglyceridemia and hypercholesterolemia.
  • Biphasic dysglycemia, characterized by initial hypoglycemia followed by hyperglycemia, was identified in rats under constant illumination.
  • Liver function was compromised, evidenced by elevated ALT and AST levels, along with hypoproteinemia and hypoalbuminemia.
  • Progressive liver damage was indicated by increased steatosis and fibrosis scores in the dark deprivation group.
  • Exogenous melatonin administration restored biochemical parameters to near-control levels and prevented liver steatosis and fibrosis.
  • Melatonin treatment increased relative liver weight and hepatocyte nuclear area, suggesting a protective effect against age-related liver changes.
