The daily rhythm of mir-142-3p can regulate its own activator Bmal1

Abstract

mir-142-3p directly targeted the 3'UTR of Bmal1 in human and mouse cells.

  • Over-expression of mir-142-3p reduced Bmal1/BMAL1 mRNA and protein levels in 293ET and NIH3T3 cells.
  • Knockdown of mir-142-3p increased Bmal1/BMAL1 mRNA and protein levels in U87MG cells.
  • The expression of mir-142-3p oscillated in serum-shocked NIH3T3 cells.
  • CLOCK/BMAL1 heterodimers directly controlled the expression of mir-142-3p in NIH3T3 cells.
  • These findings suggest a potential negative feedback loop between mir-142-3p and the core clock genes.
