Subcutaneous administration of the Cafiber/LWPLGA depot formulation resulted in a 60-fold reduction in maximum concentration compared to the immediate release formulation.

  • The extended-release formulation increased plasma half-life from 10 hours to 100 hours.
  • Sustained delivery was achieved through encapsulation of cota self-assembled fibers in a polymer depot.
  • Controlled burst release was influenced by the secondary structure and morphology of the fibers, including twisted filaments and ribbons.
  • PK modeling suggests a potential dosing frequency of every 14 days in rats while maintaining therapeutic concentration.
