Portable CRISPR tools for quick on-site detection of plant diseases

Abstract

Detection limits of 1-100 copies µL⁻¹ were achieved within 20-45 minutes using CRISPR-Cas diagnostic protocols.

  • Three field-ready assays were developed for detecting major plant pathogens: a lateral-flow test for DNA, a RT-RPA assay for RNA viruses, and an electrochemical biosensor.
  • The assays provide analytical sensitivity comparable to conventional PCR diagnostics while significantly reducing assay time.
  • Standardized workflows were created to address practical constraints such as sample inhibitors and reagent stability.
  • The protocols facilitate rapid, on-site detection of plant diseases, enhancing plant health surveillance.
