Abstract
The RPA-ECas12a platform achieves a limit of detection of 1.3 CFU/mL for Lactiplantibacillus plantarum.
- An optimal variant of crRNA, identified as 5'crRNA10, increased the catalytic efficiency of Cas12a by 33%.
- The platform can detect L. plantarum across a linear range from 10 to 10 CFU/mL.
- Detection is completed within 45 minutes and shows excellent precision with coefficients of variation less than 10%.
- High selectivity and robustness were demonstrated in tests with commercial probiotic powders and yogurts.
