Changing crRNA structure to balance reaction speeds improves one-step CRISPR-Cas12 diagnostic tests

A limit of detection of 1 copy/µL for both SARS-CoV-2 and influenza A virus was achieved within 15 minutes.

  • A one-pot detection strategy, named mutant scaffold-mediated RPA-CRISPR/Cas12a (MS-CRISPR), was developed to enhance sensitivity in molecular testing.
  • Rationally designed mutations in the crRNA scaffold help to delay the enzyme's cleavage of amplicons, preventing signal loss.
  • The method maintains a closed-tube workflow, reducing the risk of aerosol contamination during testing.
  • Clinical validation demonstrated 100% sensitivity and specificity for detecting both SARS-CoV-2 and influenza A virus in patient samples.
