CULLIN-3 regulates daily cycles of TIMELESS in the fruit fly's biological clock

Updated

Abstract

CULLIN-3 (CUL-3) is required for the circadian control of PERIOD (PER) and TIMELESS (TIM) oscillations.

  • The expression of Cul-3 RNAi or dominant negative forms of CUL-3 in clock neurons alters locomotor behavior.
  • CUL-3 deregulation induces behavioral arrhythmicity and rapidly abolishes TIM cycling under constant conditions.
  • CUL-3 has a stronger effect on TIM accumulation in the absence of PER and forms complexes with hypo-phosphorylated TIM.
  • SLMB preferentially associates with phosphorylated TIM and affects TIM more strongly in the presence of PER.
  • CUL-3 and SLMB show additive effects on TIM and PER, indicating different roles in their cycling.

What this is

  • Eukaryotic circadian clocks rely on transcriptional feedback loops, with PER and TIM proteins being crucial for this process.
  • CULLIN-3 (CUL-3) is identified as a key regulator of TIM and PER oscillations in Drosophila.
  • CUL-3 affects TIM accumulation more strongly in the absence of PER, suggesting distinct roles for CUL-3 and SLMB in regulating these proteins.

Essence

  • CUL-3 is essential for maintaining the oscillations of TIM and PER proteins in Drosophila's . Its deregulation leads to behavioral arrhythmicity and altered protein cycling, indicating its critical role in circadian rhythm regulation.

Key takeaways

  • CUL-3 downregulation disrupts behavioral rhythms in Drosophila, leading to a complete loss of TIM cycling and dampened PER oscillations. This disruption highlights the importance of CUL-3 in maintaining circadian rhythms.
  • CUL-3 interacts more strongly with hypo-phosphorylated TIM, while SLMB preferentially associates with phosphorylated TIM. This suggests that CUL-3 and SLMB have complementary roles in regulating TIM and PER stability.

Caveats

  • The study primarily focuses on the effects of CUL-3 in specific neuronal subsets, which may limit the generalizability of the findings across different neuronal contexts.
  • Behavioral analyses were conducted under controlled conditions, which may not fully replicate natural environmental influences on circadian rhythms.

Definitions

  • circadian clock: An internal biological mechanism that regulates physiological and behavioral processes in a roughly 24-hour cycle.
  • ubiquitin ligase: An enzyme that facilitates the attachment of ubiquitin to a substrate protein, marking it for degradation.
