Diabetes, diabetes medicines, and the risk of dementia: A combined review and analysis

Abstract

A total of 3,046,661 participants were analyzed in a review evaluating diabetes treatments and dementia risk.

  • Metformin, thiazolidinediones, pioglitazone, GLP1 receptor agonists (GLP1RAs), and sodium-glucose cotransporter-2 inhibitors (SGLT2is) may reduce dementia risk.
  • In the United States, metformin showed a significant effect on reducing dementia risk, while this was not observed in Eastern populations.
  • No significant reduction in dementia risk was found for dipeptidyl peptidase-4 inhibitors, α-glucosidase inhibitors, or insulin.
  • Meglitinides and sulphonylureas may be associated with an increased risk of dementia.
  • Further longitudinal studies are needed to evaluate the relative effectiveness of these treatments across different populations.
