Effects and Safety of Oral Orforglipron on Weight and Blood Sugar Control in People With and Without Diabetes

Abstract

Orforglipron led to weight reductions of more than 6 kg in patients with diabetes and nearly 12 kg in those without diabetes at the highest dose.

  • Five randomized controlled trials involving 6140 participants were analyzed.
  • Weight loss was dose-dependent, with greater reductions observed in individuals without diabetes.
  • Significant improvements were noted in body mass index, waist circumference, and glycated hemoglobin (HbA1c) levels.
  • Gastrointestinal adverse events were more common at higher doses.
  • No significant difference in acute pancreatitis incidence was found.
