Nutrient sensing at cell contact points links to GLP-1 hormone release in mouse gut cells and its role in obesity and type 2 diabetes

Abstract

ER-mitochondria interactions are dynamically induced by glucose and deoxycholic acid, increasing by 1.8- and 2.1-fold, respectively, in STC-1 cells.

  • Nutrient-induced GLP-1 secretion is linked to the dynamics of ER-mitochondria contact sites.
  • Glucose and deoxycholic acid enhance calcium exchange between the ER and mitochondria, which is crucial for hormone secretion.
  • A paracrine effect of GLP-1 influences the regulation of these contact sites in response to nutrients.
  • Disruption of organelle communication significantly impairs GLP-1 secretion from L cells.
  • Diet-induced obesity alters the ability of colonic L cells to respond to glucose, affecting GLP-1 secretion and MAM regulation.
