Abstract

Glucose significantly induced Na+ and Ca2+ signaling in STC-1 cells.

  • The release of glucagon-like peptide 1 (GLP-1) from enteroendocrine cells (EECs) may be triggered by glucose-induced calcium signaling.
  • Selective blockers of voltage-gated calcium channels, ryanodine receptors, and insulin receptors significantly reduced glucose-induced calcium signaling.
  • Blockers of TRPV4 and Na/Ca exchanger 1 (NCX1) also significantly attenuated glucose-induced calcium signaling.
  • The interaction between TRPV4 and NCX1 is essential for glucose-stimulated GLP-1 release through a specific signaling pathway involving calcium-sensitive protein kinase C alpha (PKCα).
  • A selective TRPV4 activator improved glucose tolerance in an oral glucose tolerance test in mice.
