Abstract

A fluorescence-guided approach identified diverse p21-positive senescent cell populations in aged liver.

  • Senescent cells, which no longer divide, accumulate with age and are linked to various diseases.
  • The method revealed a senescence-enriched cell population with identities including hepatocytes, macrophages, neutrophils, and plasma cells.
  • Activated macrophages and hepatic stellate cells showed a higher likelihood of being in a senescent state compared to resting cells.
  • Transcriptomic profiling of p21-expressing cells indicated varying senescence-associated secretory phenotype (SASP) programs across different cell types.
  • An aggregation of cells expressing interferon-stimulated genes was identified, with limited overlap with p21-positive cells, indicating a separate aging-related stress response.
