GLP-1 Incretin Therapy Linked to Lower Risk of Alzheimer's and Heart-Kidney Problems in Adults with Memory, Mental Health, or Sensory Risks

Abstract

In a cohort of 153,412 adults aged 50 years or older, initiation of glucagon-like peptide-1 (GLP-1) therapy was associated with a 46% lower risk of receiving a first recorded Alzheimer's disease diagnosis.

  • GLP-1 therapy initiation correlated with a reduced risk of all-cause dementia (hazard ratio 0.66).
  • A significant decrease in all-cause mortality was observed among patients starting GLP-1 therapy (HR 0.46).
  • Lower incidence rates of heart failure, cardiomyopathy, major adverse cardiovascular events, acute kidney injury, and chronic kidney disease were linked to GLP-1 treatment.
  • Negative-control outcomes showed no significant differences, suggesting the findings may be specific to GLP-1 therapy.
  • Weight-loss responders in the semaglutide group exhibited a lower 3-year cumulative incidence of Alzheimer's disease compared to non-responders.
