Links between diabetes drugs and brain growth factors in brain disorders

Abstract

GLP-1 receptor agonists (GLP-1RAs) increased brain-derived neurotrophic factor (BDNF) expression by 76% to 377% in preclinical models.

  • GLP-1RAs consistently enhanced BDNF expression and signaling in models related to diabetes, neurodegeneration, and neurotoxicity.
  • Increases in BDNF expression were associated with improved synaptic plasticity, cognition, and neuronal survival.
  • In vitro studies showed that GLP-1 and exendin-4 elevated BDNF expression and axonal transport even when exposed to harmful amyloid-beta oligomers.
  • While most neuroprotective effects aligned with BDNF upregulation, some effects occurred through alternative pathways.
