Glucagon-like Peptide-1 (GLP-1) Drugs and Thyroid Test Results: A Review Highlighting Missing Evidence

Abstract

Six studies involving 900,225 participants were reviewed to assess the effects of glucagon-like peptide-1 (GLP-1) receptor agonists on thyroid function tests.

  • Only one study directly evaluated thyroid function tests, reporting a mild decrease in free thyroxine and slight progression of thyroid nodules after 12 months.
  • Mendelian randomization analysis indicated no significant causal effect of GLP-1 receptor agonists on thyroid function.
  • Five studies focused on thyroid cancer outcomes or weight loss efficacy, without systematic evaluation of thyroid function parameters.
  • Large-scale cohort studies showed no increased risk of thyroid tumors or thyroid cancer associated with GLP-1 receptor agonist use.
  • Risk of bias assessments indicated low risk for two observational studies and moderate risk for one due to potential confounding, while randomized trials showed mixed results.
