Dexamethasone-prodrug ionizable lipids may make mRNA lipid nanoparticles less inflammatory without sacrificing delivery or editing efficiency.

Evidence

Formulation and platform experiments tested SHIELD LNPs for cytokine reduction, organ-targeted mRNA expression, base editing of EGFP and PCSK9, and repeated protein-replacement delivery.
