Melatonin Receptor Control of Reproduction: How Agomelatine Affects the Reproductive Cycle, Pregnancy, Babies, and Uterine Contractions in Rats

Abstract

Chronic agomelatine administration prolonged the gestational period and increased fetal count in pregnant rats (p<0.01).

  • Agomelatine extended the diestrus phase in non-pregnant rats (p<0.05).
  • In pregnant rats, agomelatine reduced plasma levels of oxytocin and prostaglandin-E (p<0.01).
  • In vitro studies indicated that agomelatine dose-dependently inhibited both spontaneous and oxytocin-induced uterine contractions.
  • The melatonin receptor antagonist luzindole reversed the inhibition of myometrial contractions caused by agomelatine.
  • These findings suggest a potential role for agomelatine in modulating reproductive parameters during pregnancy.
