How movement affects drug interactions with GLP-1 receptor agonists: clear evidence and a way to measure them

Updated

Abstract

Six out of eleven US prescribing-information documents show that effects on drug absorption from glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are not uniform.

  • Delayed gastric emptying from GLP-1 RAs may influence the absorption of co-administered oral medications.
  • Specific increases in exposure to drugs like thyroxine, atorvastatin, and ramipril have been documented under certain conditions.
  • Current evaluation frameworks for drug interactions may overlook motility-specific studies related to gastric emptying.
  • Total area under the concentration-time curve (AUC) may not adequately reflect the interactions of some drugs affected by gastric emptying.
  • Pharmacological heterogeneity exists among GLP-1 RAs, complicating predictions about drug absorption effects.
