Development and Testing of mRNA Vaccines Targeting the Lymphocytic Choriomeningitis Virus Glycoprotein

Updated

Abstract

Vaccination with LCMV GPC mRNA-lipid nanoparticles elicited strong GPC-specific immune responses in mice.

  • Both monovalent and bivalent vaccines demonstrated favorable formulation quality and effective antigen expression.
  • Immunization resulted in high levels of GPC-specific binding antibodies, primarily of the IgG2a subclass.
  • Pseudovirus neutralization activity was observable following vaccination.
  • Cellular immune responses were detected in all vaccinated groups, targeting shared peptide pools.
  • In a challenge model, vaccination significantly reduced disease severity and improved splenic viral clearance compared to controls.
