Abstract

Immunization with the mRNA vaccine M/N-vac in mice elicited robust Oropouche virus-specific IgG and neutralizing antibodies.

  • The M/N-vac vaccine drives the assembly of Oropouche virus-like particles with high immunogenicity.
  • Immunization induced a durable Th1-biased cellular immune response, characterized by high-level interferon-gamma secretion.
  • The vaccine produced cross-reactive antibodies against a contemporary circulating Oropouche virus strain.
  • A highly conserved potential immunodominant epitope, N2-3, was identified in the nucleocapsid protein.
  • VLP-encoding mRNA vaccine induced stronger humoral and cellular immune responses compared to a VLP protein vaccine.
