Oral GLP-1 Treatments for Obesity, Metabolic Syndrome, and Diabetes: Recent Progress, Challenges, and New Approaches

Updated

Abstract

Oral semaglutide is the first validated proof of concept for systemic peptide delivery by the gastrointestinal route.

  • The obesity-metabolic syndrome-diabetes continuum is influenced by factors such as insulin resistance and chronic inflammation.
  • Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) may provide benefits beyond glucose lowering, including weight loss and cardiovascular risk reduction.
  • Challenges such as treatment burden and access restrictions could limit the long-term effects of injectable GLP-1 RA formulations.
  • Oral GLP-1-based treatments, including new technologies for absorption and stability, are being developed to overcome barriers to oral peptide delivery.
  • Evidence suggests that oral GLP-1 RAs demonstrate significant glycemic control and weight-loss efficacy in patients with type 2 diabetes and chronic kidney disease.
