Changes in insulin-producing cell function after small intestine removal

Abstract

After 50% proximal small bowel resection, pancreatic islet hypertrophy and impaired glucose tolerance were observed in mice.

  • Hepatic steatosis and impaired glucose metabolism without insulin resistance occurred after small bowel resection.
  • The proportion of pancreatic α and β cells remained largely unchanged despite hypertrophy of islet cells.
  • Insulin secretion was significantly decreased in both static incubation and islet perfusion experiments following resection.
  • Serum levels of glucagon-like peptide-1 (GLP-1) and pancreatic GLP-1 receptor expression were reduced after resection.
  • Administration of a GLP-1 agonist mitigated some of the metabolic derangements observed after small bowel resection.
