A simple on-site test using CRISPR to detect grapevine fabavirus with improved leaf sample preparation

Abstract

The new detection method for grapevine fabavirus (GFabV) enables on-site identification within 50 minutes.

  • An optimal primer set for loop-mediated isothermal amplification (LAMP) was identified, capable of detecting all three GFabV variants.
  • Integration of LAMP with CRISPR/Cas12a resulted in a real-time assay that is exceptionally specific and sensitive.
  • This method is demonstrated to be ≤10,000 times more sensitive than conventional RT-PCR.
  • Novel extraction techniques were developed, including an 'Alkaline Resin method' for grapevine leaves and a simple 'branch-washing method' for grapevine branches.
  • The streamlined processes allow for effective on-site detection of GFabV without traditional RNA extraction.
