Activation of SCN neurons significantly drives wakefulness during the wake-maintenance zone (WMZ).

  • SCN neurons play a critical role in regulating arousal levels, particularly during the WMZ.
  • Chemogenetic and optogenetic stimulation of SCN neurons effectively increased wakefulness.
  • Disruption of SCN neurons led to reduced wakefulness during the dark period and loss of body temperature circadian rhythm.
  • SCN neurons connect to the dorsomedial hypothalamic nucleus (DMH), and stimulation of these connections can induce rapid arousal from sleep.
  • Presynaptic inputs to SCN neurons include the preoptic area, suggesting a complex circuit involved in arousal regulation.
