Possible Link Between Tirzepatide and Sudden Optic Nerve Damage: Evidence from Safety Reports and Search Trends

A total of 28 cases of ischemic optic neuropathy (ION) were identified for tirzepatide.

  • The event of ION associated with tirzepatide is considered rare but serious.
  • Disproportionality analyses indicated significant associations between tirzepatide and ION, with a Reporting Odds Ratio of 2.599.
  • Causality assessment using Evans criteria supported a 'probable' drug-event association.
  • Google Trends data indicated a rise in global search interest for tirzepatide, especially in regions with high obesity and type 2 diabetes prevalence.
  • The findings suggest a need for cautious clinical consideration and further investigation regarding the safety of tirzepatide.
