ContactSeek enhances the specificity of genome editors, achieving significant improvement in off-target detection.

  • The framework uses AlphaFold3-predicted contact probability to identify and rank regions affecting genome editor specificity.
  • Contact probability was more effective than three-dimensional structures in distinguishing interactions between on-target and off-target complexes.
  • Specificity-determining residues were pinpointed within contact regions, contributing to enhanced genome editing precision.
  • Targeted sequencing and other assays confirmed that the best variant outperformed existing high-fidelity adenine base editors.
