Use of a diabetes drug targeting hormone receptors and the risk of knee replacement surgery for arthritis

Abstract

GLP-1 receptor agonist use was associated with a significantly reduced long-term risk of total knee arthroplasty (TKA) in patients with knee osteoarthritis.

  • Patients with knee osteoarthritis who used GLP-1 receptor agonists showed lower cumulative incidence of TKA at 1, 3, 5, and 8 years compared to non-users.
  • The absolute risk difference for TKA at 8 years with 1 year of GLP-1 RA exposure was -2.80 percentage points.
  • Greater risk reductions were observed with newer generation agents, where 3 years of exposure to semaglutide or tirzepatide resulted in an absolute risk difference of -4.71 percentage points at 8 years.
  • Hazard ratios indicated a decreased risk of TKA associated with GLP-1 RA use, with HRs of 0.90 for 1 year and 0.72 for 3 years of exposure.
