How Variations in Key Body Clock Genes Relate to Blood Sugar and Insulin Resistance in Type 2 Diabetes

Abstract

Over 18,000 participants were involved in six studies linking circadian gene variants to impaired glycaemic control and insulin resistance in type 2 diabetes.

  • Variants in circadian clock genes may influence fasting plasma glucose and insulin resistance.
  • Specific gene interactions, such as CLOCK rs1801260 and MTNR1A rs2119882, were significantly associated with higher fasting plasma glucose and increased risk of type 2 diabetes.
  • Reduced expression of key clock genes was correlated with negative impacts on glycated hemoglobin and insulin resistance.
  • BMAL1 rs7950226 was identified as a susceptibility variant for metabolic syndrome.
  • The evidence available is observational and heterogeneous, indicating a need for prospective validation.
