Abstract
Prostaglandin (PG)F(2alpha) receptor mRNA production increased threefold after atosiban treatment in pregnant rats during labor.
- Genetic expression of PGF(2alpha) receptor and cyclooxygenase (COX)-2 increases in laboring rat myometrium.
- A relatively weak contractile response was observed in isolated uterine strips when stimulated with PGF(2alpha) during labor.
- Five days postpartum, PGF(2alpha) receptor mRNA levels were higher than during labor, while COX-2 mRNA returned to preparturient levels.
- Atosiban treatment in pregnant rats led to a 2.5-fold increase in maximal contractility of uterine strips stimulated with PGF(2alpha).
- Oxytocin treatment in nonpregnant rats reduced contractile responsiveness to PGF(2alpha) without changing receptor mRNA levels.
- The findings suggest that PGF(2alpha) receptor stimulation may compensate for oxytocin receptor blockage during labor.
