Using imaging to measure how drugs bind to incretin receptors in the body and brain

Updated

Abstract

SAR441255 achieved >70% GLP-1R occupancy in the pancreas and pituitary in vivo.

  • Two tracers demonstrated high specificity for GLP-1R and GIPR in vitro.
  • In vivo studies indicated SAR441255 led to dose-dependent GLP-1R occupancy greater than 70% in specific brain regions.
  • Tirzepatide exhibited lower GLP-1R occupancy compared to SAR441255.
  • SAR441255 reduced pancreatic uptake of a GIPR tracer by 23 ± 8.5%, suggesting GIPR engagement.
