Preventive semaglutide and tirzepatide treatments do not change Alzheimer's disease progression in a mouse model

Abstract

Long-acting mimetics of gut hormones did not slow Alzheimer's disease progression in 5xFAD mice over 2 or 4 months.

  • Both semaglutide and tirzepatide treatment resulted in lower body weight and improved glucose tolerance in mice.
  • There were no measurable effects on memory, learning tasks, or amyloid-β plaque deposition from either incretin therapy.
  • Chronic treatment with these drugs did not affect glial cell activation or slow neuropathological progression.
  • In a separate model, 3 days of pre-treatment with incretins did not change microglial activation or inflammatory marker levels after LPS administration.
