Single-cell RNA sequencing shows early cell behavior during stem cell therapy for large bone injuries in mice

Abstract

A combination of mesenchymal stem cells (MSCs) and a 2 mm diameter cylindrical microribbon scaffold increased the presence of mesenchymal lineage cells in a critical-size bone defect model at one week.

  • Thirteen distinct cell populations were identified in bone defect sites using single-cell RNA sequencing.
  • The proportion of immune cell types differed significantly between the scaffold only and the scaffold with MSCs groups.
  • MSCs and osteoblastic lineage cells were predominantly found in the scaffold with MSCs group.
  • Differential gene expression analysis indicated changes in immune and inflammatory cell profiles associated with MSC implantation.
  • Enhanced cell-cell interactions, particularly among MSCs, macrophages, and T cells, were observed in the group receiving MSCs.
