Tirzepatide significantly reduced body weight and white adipose tissue mass in high-fat diet-induced obese mice.

  • Tirzepatide decreased perirenal and inguinal white adipose tissue mass and reduced adipocyte hypertrophy.
  • Transcriptomic analysis indicated an upregulation of gene programs associated with thermogenesis and lipid metabolism.
  • Increased expression of browning markers, such as PGC-1α and UCP1, was observed, suggesting a shift of white adipose tissue toward a thermogenic state.
  • Tirzepatide activated the PGC-1α/UCP1 signaling pathway in cultured fat cells, promoting white fat browning.
  • The effects of Tirzepatide were reduced when cAMP signaling was inhibited, indicating its importance in mediating Tirzepatide's actions.
